If they were supposed to be people that had a natural immunity or resistance to the deadly virus, why was only one of them female? Why was there only one girl? Why wouldn't there be a roughly equal number of boys and girls in the Trial?

Yeah, for me this was about the only thing that bothered me about this movie.
I guess there are many explanations but NeoOne makes most sense if you see this as a single movie. (But it's not so I understand that some story holes and the unsatisfying ending bothers most users here.)
Otherwise I found this quite good. I especially liked the analogy about the rat lab situation, life in general.
