What are the roles that skyrocketed people from nobodies one day to A-list the next?
I was thinking of Julia Roberts. A keen observer may have already fell in love with her as an indie darling, but after Pretty Woman, everybody knew who she was. And she's a great actor who didn't go batshit crazy.
Who else?
Johnny Depp - Edward Scissorhands (He wasn't a nobody because of 21 Jump Street, but this is the breakout movie performance.)

John Travolta - Welcome Back Kotter got him started films like Saturday Night Fever and Grease made him a star. Poor choices killed it for him after that until later on when he made a comeback. Then fizzled again.
Brad Pitt - Thelma and Louise.
Sandra Bullock - Speed.
Margot Robbie - Wolf of Wall St.
Tom Hanks - Big and Forrest Gump obviously Gump moreso but I think Big played a part too.
I think the Kool-Aid Man has had the most break through performances of all. Oh Yeah.
