So, this may have been debated or discussed to death but....
Mister Mxyzptlk HAS to be the reason they would all be singing...right?
Music Meister has also been suggested as a possible instigator.
Much as I hate the character of Mxyzptlk, his powers would be a plausible explanation for getting Kara and Barry in the same universe for an episode, as well as forcing them to sing.
I want to hear Melissa Benoist sing, but I do not want to see any version of Mxy ever again. Ugh.
Or, maybe they'll just go with what most musicals do: no explanation of any kind for why people are randomly singing.

Forgot about Jeremy Jordan ( Winn) was a singer
Nearly everybody in both casts sing except Danielle Pannabaker who I have seen singing really badly in a couple of things for comedic effect (whether intentionally or not I dunno)
Melissa and Grant were on Glee
Chyler sang a few times on Greys as well as recording with her husband.
Jeremy ,Astra/Alura and Jesse Martin were all on Broadway
Carlos and Mechad are pretty good and apparently so is David Harewood
Calista can sing sort of.Laura V sings

shareThey also seem prepared to include Victor Garber who has also done musical theater and sang on Eli Stone, as well as the TV versions of Annie in 1999 and The Music Man in 2003. If they do that, I guess Franz Drameh might be included to since he's the other half of Firestorm. I don't know about his singing though.
