Whoa, good one! I think this fight would be VERY close though I could also see some equal character square offs, the roster line up would probably go something like this:
Splinter vs Shifu
Leo vs Po
Karai vs Viper (especially now that Karai is a snake herself)
April vs Tigress
Mikey vs Monkey
Raph vs Mantis
Donnie vs Crane
Shredder vs Tai Lung
This just leaves Casey who has no martial arts skills so he couldn't get matched up with anyone. Hard to tell who would win among everyone else although being the Dragon Warrior may give Po an edge over Leo. This would make an awesome cross over too, time to start writing in to Nick. 😀
A crossover would be cool if they can make it happen.

Raph vs Mantis
I'd swap these two, after all both Tigress and Raph have rage issues.
And I wonder who Master Oogway could fight, since he's a turtle.
