The creature Roland battles in the dark forest is clearly a portal guardian, but it doesn't appear to be Shardik, the bear that appeared in the Dark Tower books. I've re-watched these clips several times, and based on the anatomy of the creature (including its long tail and rodent-like teeth), I believe it's a rat. This fits in with Dark Tower lore, as one of the guardians of the Dark Tower's beams is a rat. This further confirms the theory that this will be a new journey for Roland—and this time, they'll follow the Beam of the Rat, Way of the Fish.

Fans of the Dark Tower series will know that the portal guardians tend to be named after famous animals in literature or mythology. For example, the Bear guardian was named "Shardik" after the 1974 fantasy novel by Richard Adams. I'm sure the filmmakers will throw us a bit of fanservice by also letting us know the name of this monstrous rat. For this particular guardian, there are several good pop culture candidates it could be named after: Nicodemus from Mrs. Frisby and the Rats of NIMH, Templeton from Charlotte's Web, or even Splinter from Teenage Mutant Ninja Turtles. Personally, I'm rooting for Nicodemus, but given that the rat in the trailer has gigantic spikes protruding from its back, Splinter may be more appropriate.
Now that would be very interesting. A warm up for Shardik, maybe?
shareok but, correct me if im wrong, beam guardians dont protect portals. i don't remember there being a portal when they fight shardik.
i think this just might be a simple portal guardian, like the one at dutch hill.
it cant be the rat. the beam of the rat would lead them in a completely different direction, which means no lud, no calla bryn sturgis, no devor toi. and from what i saw in the trailer and the akiva script devor toi is definitely in this movie.
i don't remember there being a portal when they fight shardik.

im on board with this being the rat, i just hope that when they return to midworld they dont follow the rat-fish beam.
the horn of eld already gives the filmmakers carte blanche, but by not following bear-turtle beam we wont see any locations familiar to us from the books.

There was no portal as in a kind of spacetime "doorway" that would allow them to instantly travel from one point to another. Shardik, like all the other Guardians, protected the machinery that a Beam is broadcast from/moored at.

I think it is a Low Man. For one, in another part of the trailer, you get to see a man "adjusting his skin"(wearing skin was something Low Men did to blend in) and there is a few seconds shot of what looks like an explosion happening in a kitchen(which will probably end up being the Dixie Pig, a restaurant where Low Men gather in NYC).

One hiccup with your theory: the Low Men are humanoid, despite possessing rodentian features. What we saw is quadrupedal and closer to a regular animal scaled up in size.
The only way I can see that monster being a Low Man is if it is some sort of evolutionary ancestor who's offspring later experienced a change in morphology due to fantastic evolution or the introduction of magical influence to their bloodline (through deliberate experimentation or accidental exposure). It MIGHT be a Low Man that was mutated like the Slow Mutants under the mountain, but I find that theory to be rather far-fetched.

Well, here's a few possibilities:
One, what is stopping ANY humanoid, rat or otherwise, from walking on all fours?
Or, two, they are obviously taking liberties with the story as is so who's to say whether or not a cinematic version of a Low Man could possibly transform into a more obvious rat-like creature than has already been portrayed?

Ok, here's one more thought. I was looking through the cast list again on the main page and several vampire characters are listed. Bats are somewhat similar to rats so maybe its a more animalistic form of the vampires in this?
OR(and I just remembered about this story), perhaps the rat creatures from "Graveyard Shift" have somehow found a nesting ground in NYC? Seems appropriate since King himself has a penchant for making connections between all of his other work and the DT series so why not carry that concept over into a cinema adaptation? Man, now I want to dig out my copy of "Night Shift" and reread it. lol
