Well, on Pee-Wee's Playhouse, he looked up Miss Yvonne and other female characters' skirts a number of times, as well as making sexual innuendos with them.

So no, he's not gay. He's just an immature man-child.

If you go on YouTube, you can watch a number of early talk show appearances from the mid to late 80s. There's one where he explains the origins of the character to Letterman.
