Trainspotting by Danny Boyle deals with illicit drugs; but there is a substantial amount of discourse on the usage of perscription drugs, particularly to get off the illicit substances (it is a good contrast/dichotomy between Prozac Nation). Like carolina-velho stated, Requiem for a Dream is another good one (closer to Trainspotting, however).
