I thought it was cancer. I think Mona mentioned it in the pilot episode when she was telling Angela why she had to hire Tony- giving her the background story.
That was back in the day when every other sitcom seemed to have a mother that had died. (Full House was another example.)

Footnote: On the t.v. show Full House. D.J. had mentioned that her mother was killed by a drunk driver.
I don't remember it being exactly said. They implied that she became ill and died. Cancer is one of the obvious choices but I don't believe they ever pinned it down.
