Latin men are often extremely protective of their families, especially their sisters. Plus he was trying to preserve the only innocent and good thing in his life. I didn't detect ANY incestuous feelings at all. Because it is obvious that Tony would never have killed Manolo if he knew beforehand that he had actually married Gina. It seems like he would have been fine with it, so long as Manolo respected her by making her his wife.
