Why didn’t he just lie to Roy and say “I’ll see what I can do” and then perhaps de-activate him on the operating table, for someone who’s supposed to be a genius he wasn’t that smart in that scene, or was he that much of an ego-maniac he thought his creation wouldn’t turn on him and realised too late just how much danger he was in?

Being a scientific genius who could design the brains and nervous systems of artificial humans, doesn't mean for an instant that he couldn't also be too arrogant and detached from ordinary humanity to understand how people react emotionally, or that a being like Roy Batty might lash out in anger when told there's no hope for him.
shareThat’s my thinking too, he just saw Roy as a piece of machinery and he must have thought that a replicant that he created wouldn’t have the programming to turn on its owner, his “more human than human” motto came back to bite him on his ass

shareThe impression I get is that Tyrell figured he was doomed the moment Roy was able to enter his bedroom without any security being alerted.
Roy wanted to know if there was a means of extending replicant lifespans. I doubt he expected Tyrell to actually help extend them, and even if Tyrell did offer to help, Roy had no reason to trust him.
