Because their eyes are very sensitive to light, their pupils contract to points ("miosis") even in very low-light situations, such as when they take off their glasses.
The don't necessarily have larger than normal pupils in dark environments (we never see that), they are just very, very sensitive to light. That's called photophobia and it is a common neurological symptom.
Their night vision may be no better than Neville's, but they can't stand the light.
They assume that Neville can't see in the dark because he only comes out in the day, but that's because he has an advantage during the day.
The Family may also just be better "dark adapted" than Neville. It takes about 30 minutes to become fully "dark adapted" - so, during that time, Neville would be more vulnerable.
